RESUMEN

PURPOSE: Patients with seasonal allergic rhinoconjunctivitis (SARC) might seek evaluation and treatment when symptoms appear during the pollen season. It is unclear whether coseasonal-initiated sublingual immunotherapy (SLIT) would be effective and safe for SARC. This study aims to identify the feasibility of initiating Artemisia annua SLIT during the pollen season. MATERIALS AND METHODS: Sixty patients with Artemisia-induced SARC were equally recruited into the SLIT and control groups during the pollen season in 2021. The SLIT group was treated with standardized Artemisia annua SLIT drops using a modified dosing schedule combined with pharmacotherapy, while the control group only received pharmacotherapy. Diary cards for clinical symptoms, rescue medication use, and adverse events (AEs) were recorded during the pollen seasons. Objective measures, including average daily combined scores of medication and rhinoconjunctivitis symptoms (CSMRS), total rhinoconjunctivitis symptom score (TRSS), total medication score (TMS), and the score of visual analog scale (VAS) were calculated to evaluate the efficacy of SLIT. Safety was assessed through the occurrence and severity of AEs. RESULTS: In total, 80.0 % (24/30) patients in the SLIT group and 86.67 % (26/30) patients in the control group completed the study. The severity of SARC, which was assessed by objective measures including CSMRS, TRSS, TMS, and VAS of the SLIT group and the control group, was generally at the same level during the 2021 pollen season, except for the medical consumption, which the score of TMS was slightly higher in the SLIT group. After one year of treatment, the scores of CSMRS, TRSS, and VAS in the SLIT group were significantly improved compared with the control group (all P < 0.001), and the difference in the TMS between the two groups disappeared (P > 0.05). Moreover, clinical improvement of the four objective measures was also observed in the SLIT group compared with the baseline value (P < 0.001). Overall, 9/24 patients in the SLIT group experienced mild local AEs, and two patients experienced mild systemic AEs during the SLIT period. CONCLUSIONS: This controlled preliminary study identified that coseasonal-initiated Artemisia annua SLIT treatment for one year was generally safe and effective in improving the symptoms of SARC patients induced by Artemisia annua pollen.

RESUMEN

This paper investigates prefabricated utility tunnels composed of composite slabs with a spiral stirrup-constrained connection, considering material nonlinearity with concrete damage. An experiment was set up based on the prototype of a practical utility tunnel project, and the results were compared with finite element (FEM) simulation results with reasonable agreement obtained. The parametric analysis was carried out considering variations of seam location, haunch height and reinforcement, and embedment depth, using FEM simulations. It is found that, as with the increase in seam distance above haunch, the load capacity increases slightly, while the ductility does not vary much. The haunch height is not found to have an apparent effect on stiffness, load capacity or ductility. The increase in the embedment depth can enhance both the yield and peak loads while decreasing the ductility. A simplified method is proposed for evaluating the seismic performance in terms of deformation coefficient considering ductility demand, based on three different methods for calculating interaction coefficients considering soil-structure interactions. The findings from this investigation provide theoretical and practical guidance for underground engineering design of prefabricated utility tunnels.

RESUMEN

The study aimed to explore the effects of local anesthetic bupivacaine on bladder cancer cells in vivo and in vitro. The cytotoxicity was detected by MTT assay. Apoptosis was measured by Hoechst 33342 staining and TUNEL. The contents of Fe2+, Malondialdehyde (MDA), Glutathione (GSH) and reactive oxygen species (ROS) were evaluated by the corresponding kit. Mitochondrial membrane potential was assessed by JC-1 kit. HE staining, TUNEL and immunohistochemistry were used to detect the xenografted tumors. Protein expression was estimated by Western blot. Bupivacaine significantly inhibited the activity of T24 cells and 5637 cells at 0.25-16 mM. Bupivacaine promoted cell apoptosis with increased concentration. bupivacaine inhibited the expression of Bcl-2 and increased the expression of Bax and cytochrome C. Moreover, bupivacaine amplified the level of Fe2+ and ROS, and restrained the expression of cystine/glutamic acid reverse transporter (xCT) and glutathione peroxidase 4 (GPX4). Further results showed that bupivacaine decreased mitochondrial membrane potential, reduced GSH, and increased MDA levels. Besides, bupivacaine attenuated the phosphorylation of PI3K, Akt, and mTOR. In addition, bupivacaine suppressed the growth of xenografted tumors, induced apoptosis and ferroptosis, and inhibited the activity of PI3K/AKT signaling pathway in xenografted tumors. Bupivacaine could induce apoptosis and ferroptosis by inhibiting PI3K/Akt signaling pathway in bladder cancer cells.

RESUMEN

Solid waste generated from building renovation, called "renovation waste" in this study, represents a major waste management problem. A particular difficulty is sourcing renovation waste arising sporadically from discrete sites for central processing. This can be characterized as a "last mile" problem in renovation waste management (RWM). This study reports good practice for dealing with the RWM last mile problem in a city in China. We conduct qualitative research comprising site investigations and interviews, organized in an in-depth case study. We discover that the city effectively solved the last mile problem by developing a multi-layer, nested waste management system, empowered further by various smart technologies and concerted collaboration from multi-stakeholders coordinated by a determined government. Nevertheless, the longevity of the RWM is contingent on confronting several challenges, including (a) achieving cost and benefit balance, (b) defining clearer standards and policies, and (c) raising stakeholders' awareness of waste management. A general RWM strategy is recommended to establish fluent channels through which to source and qualify renovation waste for central treatment. The study delves into the much neglected world of RWM and provides a valuable reference for tackling similar problems.

RESUMEN

Many seniors live in old residential communities (ORCs) with low-quality outdoor environment (OE), which hinders the residents' outdoor daily activities (ODAs). This paper empirically investigates the association of OE on ODAs for seniors living in ORCs. A questionnaire was designed and distributed in six central districts of Nanjing city. A total of 258 questionnaires was finally collected, of which 60.08%, 29.46%, 9.69%, and 0.78% respondents were scattered into four age groups (61-69, 70-79, 80-89, and ≥90), respectively. Based on reliability analysis, correlation analysis, and regression analysis, the results show that: (1) social activities are mainly associated with noise; (2) leisure activities are significantly associated with road accessibility, slip-resistance measures, greenery, and staff; (3) utilitarian-type activities are significantly associated with stairway accessibility, slip-resistance measures, greenery, and seating; (4) there is a significant association between nature-exposure activities and layout, greenery, and poor air quality. The findings could guide Chinese officials when renewing ORCs by addressing the most important outdoor environmental factors associated with ODAs.

RESUMEN

Background and purpose: A high risk of brain injury has been reported with the usage of general anesthetics such as propofol in infants. Experimental data indicated that oxidative stress and inflammation are involved in the neurotoxicity induced by propofol. Febuxostat is a novel anti-gout agent recently reported to exert an anti-inflammatory effect. The present study aims to investigate the protective property of febuxostat against the cytotoxicity of propofol in brain endothelial cells as well as the underlying preliminary mechanism. Methods: The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was utilized to screen the optimized incubation concentration of febuxostat. bEnd.3 brain endothelial cells were stimulated with 2% propofol in the presence or absence of febuxostat (10, 20 µM) for 24 h. The lactate dehydrogenase (LDH) release assay was conducted to detect cytotoxicity. The reactive oxygen species (ROS) levels were evaluated using dichloro-dihydro-fluorescein diacetate (DCFH-DA) staining, and the concentration of reduced glutathione (GSH) was determined using a commercial kit. The expressions of TNF-α, IL-6, IL-12, CXCL-1, PDPN, CXCL8, VCAM-1, and E-selectin were determined using a quantitative real-time polymerase chain reaction (qRT-PCR) and an enzyme-linked immunosorbent assay (ELISA). Western blot and qRT-PCR were utilized to determine the expressions of COX-2 and KLF6. The production of PGE2 was evaluated by ELISA. Results: First, increased LDH release induced by propofol was significantly suppressed by febuxostat. The oxidative stress (elevated ROS levels and decreased GSH level) induced by propofol was alleviated by febuxostat. Second, the upregulated inflammatory factors (TNF-α, IL-6, and IL-12), pro-inflammatory chemokines (CXCL-1, PDPN, and CXCL8), adhesion molecules (VCAM-1 and E-selectin), and inflammatory mediators (COX-2 and PGE2) induced by propofol were greatly downregulated by febuxostat. Lastly, the expression of KLF6 was significantly suppressed by propofol but greatly elevated by febuxostat. Conclusion: Febuxostat prevented the cytotoxicity of propofol in brain endothelial cells by alleviating oxidative stress and inflammatory response through KLF6.

RESUMEN

Due to the rapid pace of urbanization in China, there has been a significant increase in construction work, which has resulted in the generation of more waste. Reducing the waste at source is the most efficient way to reduce its negative impacts, and prefabrication is a construction method that does exactly that. Since prefabricated construction generates less waste compared to conventional cast-in-situ construction, it is being promoted by the Chinese government. This study investigates the benefits of prefabrication and quantifies the percentage of construction waste reduction through its application in China. It does so by using a 26-storey concrete-brick residential building as a case study, and by conducting uncertainty analysis with Oracle Crystal Ball simulation software to assess the reduction of waste when using prefabricated components in place of cast-in-situ elements. Simulation results demonstrated that the waste generation rate for in-situ timber formwork and masonry work was 10.52 and 4.77 kg/m2 respectively, and that the use of prefabricated components reduced those figures by 36.04% and 25.53% respectively. This study quantifies the benefits of prefabrication as a method for reducing the generation of construction waste in China. Not only would extensive use of prefabrication decrease the cost related to construction waste management in China, but it could also mitigate the environmental and social impacts of construction waste globally.

RESUMEN

Hexavalent chromium [Cr(vi)] which is a kind of heavy metal with strong oxidizing ability can induce cardiovascular disease (CVD), while taxifolin can protect cells and organisms against suffering from oxidative stress. In this study, the inhibitory effects of taxifolin against Cr(vi)-induced cell damage in human umbilical vein endothelial cells (HUVECs) and THP-1 cells were investigated. Cr(vi) could increase the phosphorylation of p38 and JNK, regulate the expression of Bax and Bcl-2 in both cell lines. Meanwhile, the Cr(vi) stimulation led to an increase of the expression of ICAM-1 and VCAM-1, and upregulated the adhesion of THP-1 cells to HUVECs. Furthermore, Cr(vi) could induce the activation of the nuclear factor kappa B (NF-κB) signaling pathway, the accumulation of p65 in the nucleus, and the increase in the phosphorylation of IκB and the expression of cleaved caspase-1 and IL-1ß in THP-1 cells. However, taxifolin could reverse the effects by inhibiting the activation of mitogen-activated protein kinases (MAPKs) and NF-κB signaling pathways, regulating the expression of apoptosis-related proteins, and alleviating the adhesion of THP-1 cells to HUVECs. Our findings demonstrated that taxifolin was a potential agent to prevent endothelial dysfunction, monocyte inflammation and cell adhesion induced by Cr(vi).

RESUMEN

Amidst the global trend of advocating a circular economy, various nations and regions in recent years have started to explore innovative procurement models (e.g., Public Private Partnership [PPP]) in dealing with beset issues related to construction and demolition (C&D) waste. However, PPP is suffering from problems such as 'long negotiation time', 'lack of transparency', and 'uneven risk and return allocation', which in turn lead to 'ineffective delivery' and 'poor value for money'. Using a case study, this paper reports some lessons learnt from innovative practices of procuring C&D management services in Suzhou, China. It is discovered that the public and private sectors, without prior knowledge, are operating based on a general concession framework instead of negotiating a clear-cut agreement from the outset. Several key arrangements, such as price, concession period, and strategic operations, are based on relational contract-type of agreements, which are found particularly innovative for shortening the negotiation time, fostering the trust between the relevant parties, dealing with emerging ad hoc problems, and allowing sustainable development of the circular economy. To make such PPP work, it is critical to devise institutions to prevent corruption and opportunistic behaviors. This research provides useful references to developing a circular economy. Although they focus on C&D waste management, the research findings can be applied to other public procurement settings, such as municipal solid waste management service.
